Joseph Kemnitz (UW) - Glucoregulation in CR Monkeys
---------------------------------------------------
Glucose tolerance test dose done by weight in monkeys - unlike Luigi's human
CR study.
CR monkey's show reduced fasting insulin levels, and reduce insulin
secretion, but higher insulin sensitivity, so glucose disposal still pretty
good. Glucose remains lower than AL control when given equivalent (by body
weight) challenges.
Michael Rae asked about CRonies OGTT not being very good.
Luigi Fontana clarified "not very good", said fasting insulin very low on
CRonies OGTT, and 2h response not very good - glucose doesn't go very
quickly. He'll talk more about it tomorrow. But he said it did not appear
to be due to hyposecretion of insulin, although as my first phase insulin
response (and Paul's and Khurram's) was slow and relatively low compared
with controls.
Michael Rae suggested our first phase insulin response may be attenuated by
conditioning - we never have a big glucose challenge, and the sweets we
consume don't have glucose associated with them (so habituation of cephalic
insulin response) so our pancreas has downregulated insulin release. Dr.
Kemnitz said this was a reasonable hypothesis, but he wasn't sure.
EOD feeding in rodents results in increased glycogen stores - as a reserve.
More on that in a later presentation.
